ABSTRACT.
The whitefly-transmitted Tomato chlorosis virus (ToCV) (genus Crinivirus) is associated with yield and quality losses in field and greenhouse-grown tomatoes (Solanum lycopersicum) in South America. Therefore, the search for sources of ToCV resistance/tolerance is a major breeding priority for this region. A germplasm of 33 Solanum (Lycopersicon) accessions (comprising cultivated and wild species) was evaluated for ToCV reaction in multi-year assays conducted under natural and experimental whitefly vector exposure in Uruguay and Brazil. Reaction to ToCV was assessed employing a symptom severity scale and systemic virus infection was evaluated via RT-PCR and/or molecular hybridization assays. A subgroup of accessions was also evaluated for whitefly reaction in two free-choice bioassays carried out in Uruguay (with Trialeurodes vaporariorum) and Brazil (with Bemisia tabaci Middle-East-Asia-Minor1?MEAM1 = biotype B). The most stable sources of ToCV tolerance were identified in Solanum habrochaites PI 127827 (mild symptoms and low viral titers) and S. lycopersicum ?LT05? (mild symptoms but with high viral titers). These two accessions were efficiently colonized by both whitefly species, thus excluding the potential involvement of vector-resistance mechanisms. Other promising breeding sources were Solanum peruvianum (sensu lato) ?CGO 6711? (mild symptoms and low virus titers), Solanum chilense LA1967 (mild symptoms, but with high levels of B. tabaci MEAM1 oviposition) and Solanum pennellii LA0716 (intermediate symptoms and low level of B. tabaci MEAM1 oviposition). Additional studies are necessary to elucidate the genetic basis of the tolerance/resistance identified in this set of Solanum (Lycopersicon) accessions.

Los galactomananos constituyen una valiosa fuente renovable de polisacáridos de alto peso molecular que se utilizan en la industria farmacéutica y alimentaria. Tienen propie-dades funcionales, como ser espesantes, co-gelificantes y estabilizantes de emulsio-nes. Presentan solubilidad en agua, son polisacáridos no inónicos y se extraen del en-dospermo de las semillas de leguminosas, donde cumplen funciones de hidratación y reserva. Las variedades más utilizadas a nivel comercial actualmente son Cyamopsis tetragonoloba(goma guar), Ceratonia siliqua(goma de algarrobo) y la Trigonella-foenum-graecum(goma de fenureco). Como parte del Programa de Bioprospección del bosque nativo de INIA se ha aislado y caracterizado el galactomanano presente en las semillas de Prosopis affinis (Leguminosae, Mimosaceae).Se colectaron y secaron vainas maduras de Prosopis affinis; sus semillas fueron sepa-radas manualmente (rendimiento = 17.2 ± 0.3%). El endospermo fue aislado por hincha-miento de la semilla en agua a 100ºC, y posterior separación manual (rendimiento = 3.2%, base seca). Los galactomananos se obtuvieron por extracción del endospermo en agua a temperatura ambiente y a 80 ºC, y se purificaron por precipitación con isopro-panol desde la solución acuosa (rendimiento total = 64 ± 1 %). La composición en mo-nosacáridos de los productos se analizó por TLC luego de la hidrólisis total con TFA 2N, mostrando la presencia exclusivamente de galactosa y manosa. La relación Man:Gal de las gomas (temperatura ambiente y 80 ºC) determinada por HPLC?IR fue de 1.4:1 y 1.5:1 respectivamente.Se obtuvieron los espectros NMR en un espectrómetro Bruker AVANCE III 500. La estructura de los galactomananos determinados por 1D-TOCSY y 2D-HSQC-TOCSY fue principalmente mananos lineales parcialmente sustituídos por re-siduos de galactopiranosa.La distribución molar molecular de ambas gomas fue determinada por SEC?HPLC/MALS/IR (Wyatt Dawn 8+) en una columna Agilent SEC-5, mostrando un tamaño molecular promedio de 4.8 a 5.8 x105 Da, utilizando estándares de pululanos como ca-libradores. En resumen, la estructura y distribución molecular de los galactomananos extraídos indican que poseen propiedades similares a otros productos comerciales como la goma guar y garrofín, siendo prometedores como suministro alternativo de hi-drocoloides para la industria alimentaria.
